目的 探討TRIM32過表達對MCF-7乳腺癌細胞增殖、浸潤和侵襲的影響。方法 應用Western blot、CCK-8、Transwell和wound healing實驗分析TRIM32過表達對MCF-7細胞增殖、浸潤和侵襲影響。結果 Western blot實驗顯示,TRIM32在MCF-10A正常乳腺上皮細胞中低表達,而在MCF-7乳腺癌細胞中高表達(P<0.05);與空白對照組比較,MCF-7細胞中轉染TRIM32質粒后TRIM32呈過表達(P<0.05);CCK-8實驗顯示,與空白對照組比較,TRIM32過表達后MCF-7細胞增殖率提高(P<0.05)。Transwell實驗顯示,與空白對照組比較,過表達TRIM32后MCF-7乳腺癌細胞穿孔數目增加(P<0.05);Wound healing實驗顯示,與空白對照組比較,過表達TRIM32后MCF-7乳腺癌細胞12、24 h的遷移距離增加(P<0.05)。結論 TRIM32在乳腺癌細胞中高表達,過表達TRIM32促進乳腺癌細胞增殖、浸潤和侵襲能力,其可能成為乳腺癌治療的新靶點。
Abstract:
Objective To investigate the effect of TRIM32 overexpression on the proliferation, infiltration and invasion of MCF-7 breast cancer cells. Methods Western blot, CCK-8, Transwell and wound healing experiments were used to analyze the effects of TRIM32 overexpression on MCF-7 cell proliferation, infiltration and invasion.Results Western blot experiments showed that TRIM32 was low-expressed in MCF-10A normal breast epithelial cells and high-expressed in MCF-7 breast cancer cells(P<0.05). Compared with the blank control group, MCF-7 cells were transfected with TRIM32 plasmid TRIM32 was over-expressed (P<0.05); CCK-8 experiments showed that compared with the blank control group, the proliferation rate of MCF-7 cells increased after TRIM32 over-expression (P<0.05). Transwell experiments showed that MCF-7 breast cancer cell perforations increased after overexpression of TRIM32 compared with the blank control group(P<0.05); Wound healing experiments showed that compared with the blank control group, MCF-7 breast cancer cells overexpressed TRIM32 migration distance at 12 and 24 h increased(P<0.05).Conclusion TRIM32 is highly expressed in breast cancer cells, and overexpression of TRIM32 promotes breast cancer cell proliferation, infiltration and invasion ability, which may become a new target for breast cancer treatment.
